The hit and run driver ploughed through a group of six pedestrians early on Sunday morning during a brawl outside a Hookah bar.
One person died at the scene, while a further five people were injured in the incident which police are treating as a deliberate attack.
One of the victims is reportedly in a critical condition, while the other four are believed to be stable.
The hit and run took place at 129th Street and Liberty Avenue, in the Richmond Hill neighbourhood. Two men were stabbed nearby. They both walked to a hospital where they were treated for their injuries.
Police have described the suspect as an adult male with a ponytail.
He is believed to be driving a white Hyundai Sonata.
The victim was identified by his family as 22-year-old Ricardo Chatergood.
His father Sundar said at the scene: “Oh my god, that’s my son. That’s his green sweater; those are his black sneakers. Oh my god that’s him.”
He told the New York Post: “No one knew where my son was. We could not find him anywhere.
“We have not heard from him. That’s why we came here.”
